Résumé

Flavan-3-ols, flavanones and flavones vegetal polyphenols in vitro comparative study and relation between antiproliferative effect and molecular structure. ABSTRACT: Introduction: Molecular structure is a very important factor in relation of any substances with cells or biological system. Objective: Present study is an in vitro research about the viability and proliferation activity of following products and extracts: grape seed extract (flavan-3-ols catechins polymers), cocoa extract (flavan-3-ol epicatechins), grapefruit extract (flavanones glycosides naringin y neohesperidin), skin lemon extract (flavanone glycoside eriocitrin), naringenin (flavanone), apigenin (flavone). Also study the influence of these extracts on cellular cycle and apoptosis on Vero and CaCo2. Material and Methods: We have study the different extracts and products effects concentrations on Vero cells proliferation and CaCo-2 cells culture, using the MTT assay. It together with the knowledge of the chromatographic composition of every extract of the experiment allows us to find relations between structures or molecular skeletons and his effect on the cellular proliferation. Results: In our study conditions, we have not negative effect on cellular proliferation with all the extracts, on concentrations lower than 10 µMa on Vero. The order in antiproliferative capacity could be: flavan-3-ol > flavanone > flavone. On CaCo-2, all the extracts showed antiproliferative effect or citotoxicity especially apigenine. On Vero apigenine and grape fruit seed, showed the best arrest in cell cycle. On CaCo-2, citrus and cocoa were he most significant. Cellular population percentage analyses, in apoptosis were very low regarding the total population in Vero. But, on CaCo-2, we considered relevant results on Apigenine, which showed an important apoptotic capacity after 72 hours. Conclusion: We have found the more complex molecular structures it provokes antiproliferative activity increase. This fact would interact in relationship between cell and environment.
